General Information
Osteoprotegerin (OPG) is a secreted tumor necrosis factor receptor homologue that inhibits osteoclastogenesis and increases bone density in vivo. OPG contains 4 cysteine-rich repeats and 2 ‘death domains’ (DD). OPG binds to RANKL (TRANCE) and with a similar affinity to TRAIL.

General Information
RANK ligand (RANKL), also called TRANCE, OPGL and ODF, is a member of the TNF family that is expressed in activated T cells, lymph nodes and in stromal cell lines. RANKL (TRANCE) interacts with its receptor RANK expressed on mature dendritic cells (DC) and mature osteoclasts, leading to the inhibition of apoptosis, probably through the upregulation of bcl-x. RANKL is, therefore, a useful tool for enhancing DC and osteoclast survival and activity. Osteoprotegerin (OPG, see Prod. No. ALX-522-007) is a potent inhibitor of RANKL.

General Information
The receptor activator of NF-κB (RANK) is a member of the tumor necrosis factor receptor (TNF-R) family. RANK is a type I transmembrane receptor. It is 616 aa long and contains 4 cysteine-rich motifs in its extracellular domain, while its long cytoplasmic domain spans 383 aa. RANK is mainly expressed on dendritic cells. After binding its ligand RANKL, RANK interacts with TNF receptor-associated factor 1 (TRAF1), TRAF2, TRAF3, TRAF5 and TRAF6 which mediate activation of nuclear factor NF-κB and JNK. RANK-RANKL interactions promote survival of dendritic cells as well as differentiation in osteoclasts.

The RGMC (also called Hemojuvelin) is an GPI-anchored protein involved in the iron metabolism through regulation of hepcidin expression. Mutations in the gene encoding RGMC cause severe iron overload (juvenile hemochromatosis /hemochromatosis type 2A). The protein binds to neogenin and acts as co-receptor of the bone morphogenetic protein. The mature RGMc protein is proteolytically processed and glycosylated.
