General Information
Cadherins are a family of transmembrane glycoproteins that play a key role in calcium dependent cell-cell adhesion. Several members of the cadherin family have been identified so far, including E- (epithelial), P- (placental), N- (neuronal) and M- (muscle) cadherin. M-Cadherin is expressed in myoblasts and might be involved in skeletal muscle cell differentiation.

BACKGROUND/TECHNICAL INFORMATION Protocol Modifications for Western blot:
Protocol described in [1]; To improve the detection of VE cadherin, the following modifications are recommended:
Use PAb to VE-cadherin (Prod. No. ALX-210-232) diluted to 1µg/ml in PBS containing 0.01% Thimerosal (concentration must be adjusted according to the batch used). The solution can be used several times. Incubate overnight at room temperature under stirring. Rinsing: 6 x 5 minutes plus 10 minutes at room temperature.
Secondary antibody: Biotinylated goat anti-rabbit antibody (VC-BA-1000) diluted at 1:5’000 in 1% gelatin. Avoid exposure to light. Incubation 1h 30 at room temperature under stirring. Rinse in PBS 0.05% Tween 20 for 50 minutes at room temperature under stirring.
Signal amplification using Strept ABComplex/ HRP (Dako): Follow the protocol according to Dako instructions. Dilute Strept ABComplex/HRP to 1:5’000. Avoid exposure to light. Incubate 1h at room temperature while stirring. Rinse in PBS 0.05% Tween 20, 50 minutes at room temperature while stirring. Rinse rapidly in PBS.
Detection using the Supersignal Kit Pierce: Incubate the membrane 10 minutes with the solution under red light. Membrane is then packed in Plastic wrap and put in contact with the film. This method should allow for the detection of very small amounts of VE cadherin (less than 1ng/well).
