One unit is defined as the amount of enzyme that produces 1µmol p-nitroaniline or 7-amino-4-methylcoumarin per min. at 37°C in 100mM TRIS-HCl, pH 8.5, using H-Gly-Pro-pNA or H-Gly-Pro-AMC as a substrate respectively (enzyme dilution >1:500; incubation time 10-30 min.).

Inhibitors:
Very sensitive to diisopropyl fluorophosphate (DFP), less sensitive to other serine enzyme inhibitors such as phenylmethylsulfonyl fluoride (PMSF) (Prod. No. ALX-270-184) and diethyl 4-nitrophenylphosphate (E600). Most of the usual sulfhydryl reagents have little or no effect, nor do chelating agents.
