Specific cell permeable inhibitor. Potent cell permeable and metabolically stable specific inhibitor of hSIRT1 (IC₅₀=98nM in vivo / IC₅₀=38nM in vitro; compared to hSIRT2: IC₅₀=19µM and hSIRT3: IC₅₀=48µM) with no effect on human histone deacetylases (HDACs) class I and class II, nor NAD glycohydrolase (IC₅₀>100µg). Inhibits the deacetylation of p53 (IC₅₀=1µM).

General Information

Pin1, a member of the parvulin family of peptidyl-prolyl-cis-trans-isomerases (PPIases), specifically isomerizes only the phosphorylated Ser/Thr-Pro bonds in certain proteins like Bcl-2, Cdc25C, Cdc27, p53 and Tau. Binding of Pin1 to p53 stimulates the DNA binding activity and transactivation function of p53. Pin1 also regulates the expression of cyclin D1 and is strikingly overexpressed in a subset of human tumors like breast cancer and oral squamous cell carcinoma.
