Product Description
Ski [1] and SnoN [2] are oncoproteins and transcription factors that form both homodimers and heterodimers [3]. Both have the ability to transform avian fibroblasts and induce muscle differentiation [4,5]. They recognize a specific DNA binding site (GTCTAGAC) through which they repress transcription [6]. Ski and SnoN are components of a macromolecular complex containing the co-repressor N-CoR/SMRT, mSin3 and histone deacetylase. Ski and SnoN negatively regulate TGF-β signalling by recruiting this complex to Smads [7]. Interactions between Ski and Smad2, -3 and -4 blocks Smads-mediated transcriptional activation as well as TGF-β-induced growth suppression of epithelial cells, suggesting oncogenic effects of Ski derive in part through its ability to antagonize the growth-inhibitory signals initiated by members of the TGF-β family of ligands. General Information
TAG-72, a tumor-associated oncofetal antigen expressed by a wide variety of human adeno-carcinomas. This antigen is expressed by 84% of invasive ductal breast car-cinomas, and 85-90% of colon, pancreatic, gastric, esophageal, lung (non-small cell), ovarian and endometrial adenocarcinomas. It is not expressed by leukemias, lymphomas, sarcomas, mesotheliomas, melanomas or benign tu-mors. TAG-72 is also expressed on normal secretory endometrium, but not on other normal tissues.

General Information
Trop-2 (tumor-associated calcium signal transducer 2) [1,2] is a 46-48kDa cell surface glycoprotein that acts as a calcium signal transducer [3]. The molecule is expressed at high levels in human throphoblasts and overexpressed on a large variety of human carcinomas (including breast, colon, lung and prostate cancers). As an example, Trop-2 is among the most highly overexpressed genes in primary ovarian serous papillary tumors compared to normal ovarian epithelium [4] and in in colon tumors compared with normal mucosa [5]. Trop-2 is homologous to Trop-1 and both are homologous to serum IGF-II-binding proteins. The specific role of Trop-2 in normal and tumor cells is not fully defined, but accumulating evidence suggests a regulatory role in the growth of carcinoma cells. Furthermore recent data support the hypothesis that Trop-2 is involved in tumor invasion and metastasis. Indeed, TROP2 is one of the cancer-related genes that correlates with biological aggressiveness and poor prognosis of colorectal cancer [6].

General Information
α-Actinin-4 is an actin-bundling protein of ~100kDa that is associated with cell motility, endocytosis, and cancer invasion [1, 2]. The α-actinin family comprises two non-muscle isoforms (α-actinin-1 and -4) and two skeletal muscle isoforms (α-actinin-2 and -3), with α-actinin-2 being also expressed in cardiac muscle [3]. While α-actinin-4 is almost ubiquitously expressed, particularly high concentrations are found in glomeruli. On the subcellular level it is associated with actin stress fibers, but in certain cells it also localizes to the nucleus [1]. Mutations in the α-actinin-4 gene cause an autosomal-dominant form of familial focal segmental glomerulosclerosis (FSGS) [4], which is thought to result from a defect in glomerular podocyte function. A point mutation in the α-actinin-4 gene was found to generate an antigenic peptide that is recognized by autologous cytolytic T lymphocytes (CTL) on a human lung carcinoma [5]. α-actinin-4 interacts with a variety of proteins, including the ring finger protein BERP [6], the PDZ-LIM protein CLP-36 [7], the hemidesmosomal and cell-cell contact protein BP180 [8], and the tight junction protein MAGI-1 [9]. Moreover, α-actinin-4 forms a ternary complex with Ca2+ / Calmodulin-dependent protein kinase II and Densin-180, a protein of postsynaptic densities in CNS neurons [10]. Ca2+-dependent association of α-actinin-4 with E3KARP is required for Ca2+-dependent inhibition of the Na+ / H+ exchanger 3 (NHE3) [11].
