General Information
LIGHT (derived from "homologous to Lymphotoxins, exhibits Inducible expression, and competes with Herpes simplex virus (HSV) Glycoprotein D for Herpes virus entry mediator (HVEM /TR2), a receptor expressed by T lymphocytes")is a 29 kDa type 2 transmembrane protein that belongs to the TNF superfamily [1, 2]. It is expressed in granulocytes, monocytes, platelets, immature dendritic cells and activated T cells. LIGHT is active either as membrane-anchored or secreted (soluble) forms. Three receptors of the TNF superfamily: lymphotoxin receptor (LTR), herpesvirus entry mediator (HVEM), and decoy receptor 3 (DcR3) binds to the ligand. LIGHT is involved in T-cell regulation and dendritic cell maturation [3]. It triggers several signaling pathways such as NF-κB, non-canonical NF-κB, Stat3, ERK5 / BMK1 and JNK [4]. LIGHT is released from platelets and induces a proinflammatory state in vascular endothelial cells [5]. It suppress tumor formation in vivo by inducing apoptotic cell death. Embryonic stem (ES) cells are differentiated in the presence of LIGHT [6]. Mice overexpressing LIGHT spontaneously develop severe autoimmune disease. LIGHT has been reported to increase in Rhumatoid Arthritis (RA) patients and to induce a RANKL ­dependent and -independent osteoclasts formation [7].

General Information
Lymphotoxin-α (LTα, TNF-β) is a 25 kDa type II soluble protein belonging to the TNF family. It is expressed as a trimer (LTα3), or with lymphotoxin-β (LTα1β2 or LTα2β1). LTα3 interacts with 3 different members of the TNF-receptor superfamily: HVEM (Prod. No. ALX-522-017), TNF-R1 (Prod. No. ALX-522-013) and TNF-R2 (Prod. No. ALX-522-014). LTα3 is expressed by activated T and B lymphocytes and is a pro-apoptotic cytokine, a pluripotent mediator of inflammtory and immune defense mechanisms, and moreover it plays an important role in regulating the generation of the peripheral lymphoid system.

General Information
LTβR is a 75kDa type I transmembrane protein belonging to the TNF receptor family. It is 414 aa long and contains 4 cysteine-rich repeats in its extracellular domain. It is expressed on monocytes, fibroblasts, stromal and muscle cells. It is known to activate nuclear factor NF-κB, to interact with NIK and to induce cell death via TNF receptor-associated factor 3 (TRAF3). LTβR has two known ligands: LIGHT and LTα1β2. It has been shown to be involved in splenic architecture development, lymph node genesis and affinity maturation, as well as in the development of diabetes and colitis. It has been implicated in the induction of HIV replication and impairs prion replication in mice lacking follicular dendritic cells.

Product Description
Widely described antibody against α-galactose residues with broad cross-reactivity among different species. Useful for measuring the α-Gal epitope expression on cells, glycolipids and glycoproteins, characterization of hyperacute rejection (HAR) in organ and tissue transplantations and monitoring xenotransplantation experiments. The IgM isotype mimicks in vivo reactions and can be used for cytotoxicity assays for α-Gal specific pathways (addition or presence of complement necessary). Ideal as a high-throughput screening tool for inhibitors of antibody induced cytotoxicity.
Organ transplantation from pig to human result in HAR. Humans naturally produce large quantities of anti-α-Gal antibodies, which represent 1-3% of all circulating immunoglobulins and are produced by about 1% of all B cells. When pig organs or tissues are transplanted into the human body, the IgM isotype of anti-Gal binds to α-Gal epitopes, which causes activation of the complement cascade, resulting in cell lysis. This rapid activation of complement by anti-Gal IgM is an immunological barrier that poses the greatest risk of initiating HAR. The monoclonal antibody M86 to Galα1-3Gal epitopes developed by U. Galili, et al. is an essential tool in the study of human xenotransplantation and related research.
