General Information
Hrs/Hgs (hepatocyte growth factor-regulated tyrosine kinase substrate) is a tyrosine-phosphorylated member of a protein family characterized by a conserved 3´-phosphoinositide-interacting RING zinc finger domain, the so called FYVE finger. A variant of Hrs, Hrs-2 is an ATPase that has been shown to play a regulatory role in the docking and/or fusion of synaptic vesicles to plasma membranes through a calcium-regulated interaction with SNAP-25. Hrs-2 physically interacts with Eps15, a protein required for receptor-mediated endocytosis. The Hrs-2/Eps15 interaction is calcium dependent, inhibited by SNAP-25 and alpha-adaptin, and results in the inhibition of receptor-mediated endocytosis. Immunoelectron microscopy reveals Hrs-2 localization on the limiting membrane of multivesicular bodies, organelles in the endosomal pathway. Recent data suggest that Hrs-2 acts to provide communication between endo- and exocytic processes.

General Information
In 2006, T. Waerner, et al. characterized ILEI as a key player in both epithelial to mesenchymal transition and late-stage carcinoma progression. EMT is a process that involves changes in gene expression, cytoskeleton organization, cell adhesion, and extracellular matrix composition. This process is important during tissue formation and organogenesis, but also during cancer progression, metastasis and chronic inflammation. ILEI is overexpressed and/or altered in intracellular localization in multiple human tumors, an event strongly correlated to invasion/EMT, metastasis formation, and survival in human colon and breast cancer.

General Information
TGF-β receptor III (TGF-βRIII; betaglycan) is an ubiquitously expressed transmembrane proteoglycan TGF-β co-receptor [1] that regulates TGF-β signaling by increasing the binding of TGF-β to the TGF-β receptor. (Note: The TGF-β receptor III is a co-receptor of TGF-β receptor, composed of subunit I and II; TGF-β receptor III increases the binding of TGF-β to the subunit II. In this way it can regulate the signaling.) TGF-β receptor III expression is lost in many human breast and prostate cancers and correlates with the resistance to the tumor suppressor effects of TGF-β [2] . The overexpression of TGF-β receptor III in breast cancer cells suppresses their tumorigenicity in an animal model [3].
