Inhibitor of DNA methyltransferase. Restores mRNA and protein expression of caspase-8 and TRAIL (tumor necrosis factor-related apoptosis inducing ligand) sensitivity of resistant cell lines. Enhances apoptosis induced by HDAC (histone deacetylase) inhibitors.

Product Description
Cell permeable, nonsteroidal glucocorticoid receptor modulator with high binding affinity. Exerts anti-inflammatory potential by down-modulating TNF-induced pro-inflammatory gene expression, such as IL-6 and E-selectin. Unlike dexamethasone (Prod. No. ALX-370-002) it does not enhance glucocorticoid response element (GRE)-driven gene expression or induce glucocorticoid receptor (GR) binding to GRE-dependent genes in vivo.

General Information
DNA methylation is an important regulator of genetic information, shutting down gene expression through the addition of methyl groups to cytosine (C) bases in the DNA. The establishment of DNA methylation patterns requires de novo methylation that occurs predominantly during early development and gametogenesis. DNA methyltransferase-1 (Dnmt1) "maintains" C methylation after DNA replication by methylating the daughter DNA strand using the methylation pattern of the parental strand as a template. Dnmt3a and Dnmt3b are essential for the de novo methylation. They show nonoverlapping functions in development, with Dnmt3b specifically required for methylation of centromeric minor satellite repeats. Both Dnmt3a and Dnmt3b expression is elevated in several tumor cell lines comparable to Dnmt1. It has also been suggested that mutations of Dnmt3b are responsible for the ICF immunodeficiency syndrome.
