Calcium specific chelator and indicator. Upon binding calcium there is a shift in the UV spectrum and an enhancement of fluorescence. QUIN 2 was a member of the first generation of Ca
2+
chelators introduced by Tsien in 1980. These chelators have backbones similar to that of EGTA (ethylenglycol-
bis
(β-aminoethyl)-N,N,N',N'-tetraacetic acid) and, like EGTA, they are able to chelate Ca
2+
with high affinity and specifity. Moreover, these molecules contain aromatic rings that interact electronically and sterically with the chelating backbone. Thus chelation results in changes in the UV-absorbing and fluorescing properties of the molecules. The resulting spectral changes allow these substances to be used as very sensitive probes for Ca
2+
ions, Ca
2+
concentrations in single cells of various types can be measured for the first time! TPEN (Prod. No.

The DY-labels of the RedPAck are amino-reactive fluorescent biolabels designed for excitation by red lasers (He/Ne; 633nm) and diode lasers (635-650nm). They can be used for covalent coupling to proteins and other biomolecules containing primary amino groups like amino-modified DNA-oligomers and amino-modified biotin. The chromophores responsible for the absorption maxima are based on bridged hemicyanines and merocyanines.

-specific fluorophore. Can be loaded into cells via microinjection or scrape loading. Excitation and emission spectra do not undergo a shift and the sensor is essentially nonfluorescent before Ca
