Gel super shift experiments show disruption of vitamin D receptor (VDR)/RXRβ/VDRE (DNA) complex formation when RXRβ was preincubated with Prod. No. ALX-804-104, yet experiments using THR showed a supershift of the thyroid hormone receptor (THR)/RXRβ heterodimer by this product.

General Information
RIG-I (retinoic acid-inducible gene I; Ddx58), a member of the DEAD-box RNA helicases, is activated by cytoplasmic dsRNA produced during the viral replication. The protein is characterized by a N-terminal region with two caspase recruitment domains (CARD) and a C-terminal region harboring potential ATP-dependent RNA helicase activity. RIG-1 recruits the CARD adaptor inducing IFN-β (Cardif) in a CARD-CARD-dependent manner resulting in NF-κB and IRF3 activation.

General Information
Certain serine/threonine protein kinases, such as ASK1, RIP, DAP, and ZIP kinases, are mediators of apoptosis. Receptor interacting proteins including RIP and RIP2/RICK mediate apoptosis induced by TNF-R1 and Fas, two prototype members in the death receptor family. A novel member in the RIP kinase family was recently identified and designated RIP3 [1-3]. RIP3 contains an N-terminal kinase domain but, unlike RIP or RIP2, lacks the C-terminal death or CARD domain. RIP3 binds to RIP and TNF-R1, mediates TNF-R1 induced apoptosis, and attenuates RIP and TNF-R1 induced NF-κB activation. Overexpression of RIP3 induces apoptosis and NF-κB activation. The messenger RNA of RIP3 is expressed in a subset of adult tissues [2,3]. Meanwhile RIP4 has been identified and initially characterized. These recent data suggest that RIP4 is involved in NF-κB and JNK signalling and that caspase-dependent processing of RIP4 may negatively regulate NF-κB-dependent pro-survival or pro-inflammatory signals [4].

General Information
RNase L is an endoribonuclease regulated by 2-5A synthetase (2-5A). It is a 741 aa cytoplasmic and mitochondrial protein in human. RNase L functions in the antiviral and antiproliferative activities of interferons (IFN) and in multiple apoptotic pathways, standing in the first line of the innate cellular defense mechanism induced by the IFN pathway.
In its native state, the N-terminal half of RNase L functions as a repressor of the ribonuclease domain in the C-terminal half. Upon binding with high affinity to 2-5A molecules, the inactive monomeric form of RNase L is induced to dimerize into its catalytically active form.
Since mutations in RNase L relate to an increase incidence of prostate cancer, the activation of RNase L directly or indirectly suppresses prostate tumorigenesis and/or metastasis. The 2-5A/RNase L pathway is implicated in mediating apoptosis in response to viral infections and to several types of external stimuli. The fungal ribonuclease, α-sarcin, the RNA modyfing enzyme, ricin A chain, and UV light each lead to ribotoxic stress responses involving damage to the 3-end of the large ribosomal RNA. These treatments activate c-jun N-terminal kinases (JNKs). JNK activation in response to UV irradiation has been linked to apoptosis through Bax (requires caspase 3 activity, appearance of cytosolic cytochrome C indicating involvement of mitochondria).
