Product Description
Kit provides a fast and colorimetric-based method that eliminates radioactivity, extractions, or chromatography, as used in traditional assays. The new procedure requires only two easy steps, both performed in the same microtiter plate. First, the histone deacetylase (HDAC) colorimetric substrate, which comprises an acetylated lysine side chain, is incubated with a sample containing HDAC activity. Deacetylation sensitizes the substrate, so that in the second step, treatment with the lysine developer produces a chromophore. The chromophore can be easily analyzed at 400 or 405nm using an ELISA plate reader or a spectrometer. The assay is well suited for high throughput screening applications. HDAC inhibitors, substrates, and antibodies are also available separately.

General Information
HMGB1 is present in almost all mammalian nuclei where it functions in many nuclear processes including gene transcription. Post-translational methylation at lysine 42 (Lys
42
) of HMGB1 causes its cytoplasmic localization in neutrophils [1]. HMGB1 is also found extracellularly after passive release by necrotic cells or through active secretion in response to angiogenic and inflammatory signals [2, 3]. Apoptotic cells also release HMGB1 but in an oxidized form that does not trigger the immune response [4]. Secreted HMGB1 may signal through the receptor for advanced glycation end products (RAGE), and via toll-like receptors, TLR2 and TLR4 [5]. HMGB1 functions as a late mediator of immune response [3] : it is released from dying tumor cells, stimulates mDC tumor antigen processing through its interaction with TLR4 [6]. HMGB1 is overexpressed in a number of cancers where the protein contributes to the development of tumors [7].
