Apaf-1 and caspase-9 are required for cytokine withdrawal-induced apoptosis of mast cells but dispensable for their functional and clonogenic death: V.S. Marsden, et al.; Blood 107, 1872 (2006) Abstract; Full Text
General Information
Capases are a highly conserved family of cysteine proteases and are synthesized as inactive zymogens, which are cleaved at Asp-x sites during apoptosis, generating large and small subunits that heterodimerize to form the active enzyme. Caspase-1 is a member of the ICE/CED-3 family of caspases and is detected in the pro-form as a protein of 45 kDa.
Caspase-1 generates the bioactive form of the pro-inflammatory cytokine IL-1β from its biologically inactive precursor. Overexpression of caspase-1 in cells induces apoptosis, which can be inhibited by cytokine response modifier A (CrmA). Caspase-1 mRNA is predominantly expressed in the spleen and lung and to a lesser extent in skeletal muscle, kidney, testis and heart and within these tissues macrophages preferentially express caspase-1. Caspase-1 deficient mice have no gross abnormalities, however, macrophages from caspase-1-deficient mice are defective in IL-β processing and release.

Caspase-2 is not required for thymocyte or neuronal apoptosis even though cleavage of caspase-2 is dependent on both Apaf-1 and caspase-9: L.A. O'Reilly, et al.; Cell Death Differ. 9, 832 (2002), Methodology Abstract

Identification and characterization of specific DNA-binding complexes containing members of the Myc/Max/Mad network of transcriptional regulators: A. Sommer, et al.; J. Biol. Chem. 273, 6632 (1998) Abstract; Full Text
General Information
Myc proteins are important regulators of cell behaviour and work as transcription factors that belong to the basic region/helix-loop-helix/leucine zipper family of DNA binding proteins. To regulate its target genes, Myc recruits several different cofactors (e.g. PARP-10). They control a variety of cellular functions including protein synthesis, cell cycle progression, and apoptosis, thereby modulating cell growth and proliferation, differentiation, and survivial of cells. The family of myc genes includes B-myc, L-myc, N-myc and s-myc; however only c-myc, L-myc and N-myc have neoplastic potential.
The c-Myc gene was discovered as the cellular homolog of the retroviral v-myc oncogene. It is a proto-oncogene, whereas the deregulated expression of c-Myc is observed in the majority of human and animal tumors often with poor prognosis.
The proto-oncogene c-Myc is implicated in various physiological processes; cell growth, proliferation, loss of differentiation and cell death (apoptosis). It also has been implicated in the loss and dysfunction of insulin-producing beta cells in diabetes. Recent studies in mice suggest that c-Myc has unexpected functions during both self-renewal and the differentiation of stem and early progenitor cells. c-Myc activity is normally tightly controlled by external signals including growth factors, mitogens and beta-catenin which promote, and factors as TGF-beta which inhibit.

General Information
Oxygen availability plays a major role in the regulation of expression of many different genes including erythropoietin, nitric oxide synthase (NOS), heme oxygenase 1 (HO-1), glucose transporters and vascular growth factors necessary for the maintenance of homeostasis in hypoxic conditions. Hypoxia-inducible factor 1α (HIF-1α) has been identified as a bHLH-PAS family member which is instrumental in the oxygen-dependent regulation of these genes. HIF-1α rapidly accumulates in the nucleus upon exposure to hypoxic conditions where it heterodimerizes with the aryl hydrocarbon nuclear receptor translocator, ARNT, also referred to as HIF-1β.
