Product Description
Cardiotonic glycoside which presents a bufadienolide-steroid structure. Besides ouabain (Prod. No. ALX-350-066), from the class of cardenolides hellebrin is the second unique water soluble cardiotonic glycoside. Extremely potent inhibitor of Na+/K+-ATPase  (sodium pump) blocking the active efflux of Na+ and reuptake of K+ of this membrane enzyme. Shows a positive inotropic effect by increasing the intracellular Ca2+ concentration. Strong immunosuppressor inhibiting T cell activity with much higher potency than cortisol or cyclosporin A (Prod. No. ALX-380-002). Induces caspase-dependent apoptosis.

General Information
The sarcoplasmic reticulum (SR) is in part responsible for maintaining the level of intracellular Ca2+ in cardiac and skeletal muscle by storing and releasing Ca2+. Several intralumenal SR Ca2+ binding proteins have been identified, the most prominent of these is calsequestrin. Calsequestrin is a calcium binding protein known to sequester calcium accumulated in the sarcoplasmic reticulum of muscle cells during relaxation and is found discretely localized to the junctional and corbular (terminal cisternae) SR. Calsequestrin functions to localize Ca2+ near the junctional face of the terminal cisternae from which Ca2+ can be released into the cytosol via the ryanodine receptor. This protein is highly acidic and has a large capacity and moderate to low affinity for Ca2+. Two mammalian isoforms of calequestrin exist, a cardiac and a skeletal muscle isoform, which are the products of different genes and are 63% identical. This antibody recognizes both cardiac and skeletal muscle calsequestrin from a variety of species.

Product Description
Na+/Ca2+-K+ exchanger proteins (NCKX1) have been reported to occur in retinal photoreceptors and in brain (NCKX2). Unlike the abundant and widely expressed Na+/Ca2+ exchanger which catalyzes the exchange of 3 Na+ ions for 1 Ca2+ ion to modulate intracellular Ca2+ levels, NCKX isoforms exchange 4 Na+ ions for 1 Ca2+ and 1 K+ ion. The Na+/Ca2+-K+ exchanger uses the transmembrane Na+ gradient to catalyze countertransport of Ca2+ in a K+-dependent mechanism against its electrochemical gradient. It is hypothesized that NCKX2 plays a significant role in calcium homeostasis and the regulation of neuronal function.

APPLICATION: ELISA (direct and indirect: 1:2’000-1:5’000)
Western Blot (1:2’000-1:5’000 using ECL. Suggested blocking and dilution buffer is PBST containing 0.05% Tween 20 and 5% skim milk. Suggested incubation time is 1 hour at room temperature).
Optimal conditions must be determined individually for each application.
