Product Description
Inhibitor of G2 DNA damage checkpoint (IC50=8µM) and check point kinases 1 (Chk1) (IC50=3µM) and 2 (Chk2) (IC50=3.5µM) [1,3]. Unlike other checkpoint inhibitors DBH does not inhibit ataxia-telangiectasia mutated (ATM) or ATM-Rad3-related protein. Also inhibits MAP kinase kinase 1 (MEK-1) (IC50=881nM) but is not as potent as 10Z-hymenialdisine (Prod. No. ALX-350-289) (IC50=6nM) [2].

Product Description
Originally isolated from the sponges Axinella verrucosa and Acantella aurantiaca. Potent inhibitor of mitogen-activated protein kinase kinase-1 (MEK-1) (IC50=6nM). Blocks the in vivo phosphorylation of the microtubule-binding protein tau at sites that are hyperphosphorylated by glycogen synthase kinase-β (GSK-3β) and CDK5/p35 in Alzheimer’s disease. Inhibitor of DNA damage checkpoint at G2 phase (IC50=6µM), cyclin-dependent kinases CDK1/cyclin B (IC50=22nM), CDK2/cyclin A (IC50=70nM), CDK2/cyclin E (IC50=40nM), CDK4/cyclin D1 (IC50=600nM), CDK5/p25 (IC50=28nM), GSK-3β (IC50=10nM), and casein kinase 1 (CK1) (IC50=35nM). Inhibitor of NF-κB activation and of various pro-inflammatory cytokines such as IL-1, IL-2 (IC50=2.4µM), IL-6, IL-8, TNF-α (IC50=1.4µM) and nitric oxide (NO) (IC50=0.8µM) in a variety of cell lines.

General Information
Rad9 is a cell cycle checkpoint protein: DNA damage leads to extensive phosphorylation of Rad9 [4] which then inhibits progression from G2 in order to preserve cell viability, allowing proper ligation of DNA breaks before the entry of cells into mitosis [1,2]. The human homolog [3] of S. pombe Rad9 is physically proximal to the upstream (DNA damage) signal and associates with two other checkpoint control proteins, Rad1 and HHUS1 [5,6]. Rad9 also interacts with Bcl-2 family proteins Bcl-2 and Bcl-XL, but not with BAX or BAD. Overexpression of Rad9 in mammalian cells leads to apoptosis, where as antisense Rad9 RNA suppresses cell death induced by methyl methanesulphonate [7].

Product Description
Ataxia telangiectasia is a recesive childhood disease by mutations in the ATM (AT-mutated) gene. ATM protein and Ataxia telangiectasia and Rad3-related protein (ATR) belongs to a family of protein kinases that share similarities at their C-terminal region with the catalytic domain of PI(3)K (phosphoinositide 3-kinase). ATM and ATR function in response to DNA damage and repair as well as at DNA replication checkpoints during the cell cycle. ATM and ATR directly phosphorylate the breast/ovarian cancer susceptibility gene BRCA1 following DNA damage and also regulate Chk1, which is required for the G2/M checkpoint. Like DNA-PK ATR phosphorylates p53 on serine 15. ATM has also been shown to phosphorylate murine double minute 2 (MDM2) and to interact with histone deacetylase. ATM, but not ATR is cleaved during apoptosis very similar to PARP.

General Information
BACKGROUND/TECHNICAL INFORMATION Recommendations for Western blotting

ATM is a high MW protein of ~350kDa and as such, conditions may require optimization. 100-150µg of cell lysate is loaded on 5% acrylamide gels (until 200kDa marker is halfway through the gel) and transferred to nitrocellulose using semi-dry transfer at 9V constant voltage for 2 hours. Detection should be done using HRP-conjugated anti-rabbit IgG. This antibody may detect several smaller non-specific bands present in both ATM mutant and wild type cells. Therefore, careful titering of antibody concentrations is recommended.
