IMMUNOGEN: Recombinant human TNF-α (tumor necrosis factor-α).
SPECIFICITY: Recognizes human TNF-α. Does not cross-react with mouse TNF-α or human or mouse TNF-β.
APPLICATION: ELISA: Use as capture or detection antibody in sandwich ELISA.
Flow Cytometry: Suitable as primary antibody for staining in FACS analysis; recommended concentration is 10µg/ml as determined on PBMC. Method: PBMC were incubated with PMA, ionomycin, and with  monensin to inhibit secretion. Incubation for 20 minutes with TNF-D at 10µg/ml; stained with goat-anti-mouse IgG-FITC second step reagent for 15 minutes.
Functional Application: Selectively neutralizes human TNF-α bioactivity. TNF-D requires an incubation of antigen and antibody for 1 hour at 37 °C or 16 hours at 4 °C to completely neutralize TNF-α cytolytic activity.
Optimal conditions must be determined individually for each application.
