Product Description
The kit uses aracidonyl thiophosphatidylcholine (Prod. No. ALX-300-141) which serves as a substrate for all phospholipase A2s including Ca2+-dependent cytosolic PLA2 (cPLA2). Upon hydrolysis of the thioester bond, free thiols are detected using Ellmans reagent (DTNB) (Prod. No. ALX-400-034). The assay can be used to determine cPLA2 activity in purified preparations, cell cultures or tissue homogenates. Use of the assay with preparations known to contain more than one type of PLA2 will result in the measurement of total PLA2 activity, rather than cPLA2 activity alone. cPLA2 is the only arachidonic acid specific PLA2, stimulating eicosanoid release and signal transduction. This assay kit can be used for specific determination of the activity of the 85kDa form of the enzyme.
