General Information
BACKGROUND/TECHNICAL INFORMATION
MMP-3 hydrolyzes components of the extracellular matrix like proteoglycan, laminin, fibronectin, gelatin and collagen types III, IV and IX. It also activates pro-MMP-9 and pro-MMP-8 and superactivates plasmin activated MMP-1. MMP-3 is secreted as a latent proenzyme and is activated by a variety of proteinases, e.g. plasmin, trypsin, chymotrypsin, cathepsin G or human neutrophil elastase. The activating process generates an active 45kDa species which converts easily to a highly active 28kDa form. Product ALX-201-042 displays the same activity as the 28kDa form.

General Information
Caspase-9 (also known as Ice-Lap6 and Mch6) is a member of the interleukin-1
b converting enzyme (ICE) family of cysteine proteases. Similar as other caspases, caspase-9 also exists in cells as an inactive proenzyme. During the initiation of apoptosis procaspase-9 is processed at aspartate residues to form active caspase-9. As one of the initiator caspases, active caspase-9 functions to trigger the activation of downstream effector caspases, leading to disassembly of cell structures. 1x reaction buffer (final concentration), enzyme (4 reactions with 1, 2, 3 or 4µg were performed), 10µg phosphatidylinositol, 20µM cold ATP and 10µCi

General Information
Caspase-1 is the prototypical member of the caspase family of cysteine proteases. Caspase-1 exists in cells as an inactive 45kDa pro-enzyme. The pro-enzyme is matured by proteolysis to yield large (20kDa) and small (10kDa) subunits. The active enzyme is a heterotetramer consisting of two large and two small subunits. To date the mechanism of regulation of caspase-1 activation is complex and poorly understood. In THP-1 cells, a large proportion of the caspase-1 is present in the inactive pro-enzyme form.

General Information
Caspase-2 is a member of the interleukin-1β converting enzyme (ICE) family of cysteine proteases. Similar as other caspases, caspase-2 also exists in cells as an inactive proenzyme. During apoptosis, procaspase-2 is processed at aspartate residues by self-proteolysis and/or cleavage by upstream caspases. The processed form of caspase-2 consists of large (19kDa) and small (12kDa) subunits which associate to form the active enzyme. The expressed caspase-2 spontaneously undergoes auto-processing to yield the subunits characteristic of the native enzyme. The active recombinant caspase-2 is routinely tested for its ability to enzymatically cleave the caspase-2 colorimetric substrate Ac-VDVAD-pNA (Prod. No.

General Information

During apoptosis procaspase-3 is processed at aspartate residues by self-proteolysis and/or cleavage by upstream caspases, such as caspase-6, caspase-8 and granzyme B. The processed form of caspase-3 consists of large (17kDa) and small (11kDa) subunits which associate to form the active enzyme. Active caspase-3 has been shown involved in the proteolysis of several important molecules, such as poly(ADP-ribose) polymerase (PARP), the sterol regulatory element binding proteins (SREBPs), focal adhesion kinase (FAK) and others.

SPECIFIC ACTIVITY:
5’000U/mg protein. One unit is defined as the amount of enzyme that cleaves 1nmol of the caspase substrate IETD-pNA per hour at 37°C in a reaction solution containing 50mM HEPES, pH 7.2, 50mM NaCl, 0.1% CHAPS, 10mM EDTA, 5% glycerol and 10mM DTT.
APPLICATION:
Useful in screening caspase inhibitors, studying enzyme regulation, determining specificity of caspase substrates or as positive control in caspase activity assays. We recommend using 1U per assay for analyzing caspase activity.

General Information
Caspase-8 is a member of the interleukin-1β converting enzyme (ICE) family of cysteine proteases. Same as other caspases, caspase-7 also exists in cells as an inactive proenzyme. During apoptosis procaspase-8 is processed at aspartate residues by self-proteolysis and/or cleavage by another caspase. The processed form of caspase-8 consists of large and small subunits which associate to form the active enzyme.
