General Information
Procaspase-3 (human) (recombinant) is highly activatable by treatment with active human recombinant caspase-8  (Prod. No. ALX-201-062). Activation is routinely tested using 10-30ng of procaspase-3 treated with 0.-0.5 units of the active recombinant caspase-8 in 100µl of reaction buffer for 30 minutes. Activation of procaspase-3 can be monitored using the Caspase-3 Fluorometric (Prod. No. ALX-850-216) and Colorimetric Assay Kits (Prod. No. ALX-850-215).

One unit of the recombinant human caspases is defined as the enzyme activity that cleaves 1 nmol of the individual caspase substrate (YVAD-pNA for caspase-1, VDVAD-pNA for caspase-2, DEVD-pNA for caspase-3, VEID-pNA for caspase-6, DEVD-pNA for caspase-7, IETD-pNA for caspase-8, LEHD-pNA for caspase-9, and IETD-pNA for caspase-10), per hour at 37°C in a reaction solution containing 50mM HEPES, pH 7.2, 50mM sodium chloride, 0.1% CHAPS, 10mM EDTA, 5% glycerol, and 10mM DTT.
