General Information
Caspase-11 not only activates caspase-1, that is required for the maturation of proinflammatory cytokines such as interleukin-1 (IL-1) and interleukin-18 (IL-18), but also activates caspase-3, leading to cellular apoptosis under pathological conditions. In most cells, caspase-11 is only expressed upon induction with pro-inflammatory stimuli. Caspase-11 is an essential mediator of endotoxic shock and caspase-11-deficient mice are resistant to endotoxic shock. LPS-induced caspase-11 regulates lymphocyte apoptosis by activating both caspase-3 and caspase-7. Human caspase-4, thought to be the homolog of mouse caspase-11, may be an effective therapeutic target for treatment of septic shock.

General Information
Caspase-12 is a member of the ICE/CED-3 family of caspases and is detected as a protein of ~51kDa. Overexpression of caspase-12 in cells induces apoptosis. Capase-12 is localised to the endoplasmatic reticulum (ER) and activated by ER stress and specifically cleaves and activates pro-caspase-9 and is also thought to be involved in the apoptosis induced by viral infection. Caspase-12-deficient mice are resistant to ER stress-induced apoptosis. Caspase-12 is probably expressed in many and perhaps all mouse tissues; and is expressed at high levels in skeletal muscle, heart, lung and kidney, and at moderate levels in brain and liver. In many humans caspase-12 is a pseudo gene and not expressed.

General Information
Caspase-14 (MICE for mini-ICE) is a recently member of the caspase family, related to human/mouse caspase-2 and human caspase-9. It possesses all the typical amino acid residues of the caspases involved in catalysis, including the QACRG box, and contains no or only a short prodomain. In contrast to the other short prodomain caspases (caspase-3, -6, -7), caspase-14 preferentially associates with large prodomain caspases, including caspase-1, -2, - 4, -8, and -10. Caspase-14 is not processed by multiple death stimuli including activation of members of the tumor necrosis factor receptor family and expression of proapoptotic members of the bcl-2 family.

General Information
The CD14 glycoprotein, gp 55, is present on most monocytic and macrophage like cell types: monocytes, macrophages, Kupffer cells, pleural phagocytic cells and dendritic reticular cells. CD14 is also observed on granulocytes and activated or transformed B-cells. Furthermore CD14 is present in a soluble form in mouse serum, urine and other body fluids. The CD14 molecule has been reported to be a receptor for endotoxin.
BACKGROUND/TECHNICAL INFORMATION
Binding titer at mouse CD14 transfected CHO-cells is more than 20’000.
Positive control: The antibody binds to mouse macrophage like cell line J774 (50%) and to native mouse CD14 in mouse EDTA-blood after 6 hours of stimulation with LPS (70%).
