General Information
Cylindromas are benign adnexal skin tumors caused by germ line mutations in the CYLD gene. In most cases the second wild type allele is lost in tumor tissue suggesting that CYLD functions as tumor-suppressor. CYLD is a protein of 956 amino acids harboring a functional deubiquitinating domain at the C-terminal end. CYLD was shown to interact with TRAF2, NEMO and TRIP (Trompouki et al.; Brummelkamp et al.; Kovalenko et al.; Regamey et al.). Furthermore, CYLD is a negative regulator of NF-κB activation mediated by TNF family members and IL-1β. Downregulation of NF-κB activity depends on the deubiquitinating activity of CYLD which removes lysine-63 linked ubiquitin chains from target proteins. These findings indicate that cylindromas arise through constitutive NF-κB activation leading to hyperproliferation and tumor growth.

General Information
LRRK2 is a large protein with multiple domains including several ankyrin, leucine-rich, and WD40 repeats, a Ras-like small GTPase family domain named Roc, and a kinase domain that is closely related to the RIP kinase domain. LRRK2 gene is expressed in brain as well as in other tissues such as lung, liver and heart. LRRK2 might be central to the pathogenesis of several major neurodegenerative diseases associated with parkinsonism. Several dominantly inherited missense mutations in the gene encoding LRRK2 have been identified in several families that exhibit a broad spectrum of neuropathological features.

General Information
RIG-I (retinoic acid-inducible gene I; Ddx58) and Mda5 (melanoma differentiation-associated gene 5, also known as Ifih1 or Helicard) are proteins that sense viral replication intermediates, such as double-stranded RNA and triggers the host antiviral programs. These molecules signal the downstream activation of NF-κB and IFN regulatory factor (IRF) -3, which coordinately regulate the expression of type-I interferons.
Cardif (also called VISA/IPS-1/MAVS) is a new CARD (caspase activation and recruitment domain)-containing adaptor protein that interacts with the CARD domain of RIG-I and Mda5, leading to the activation of NF-κB and IRF3. Cardif is located to the mitochondrial outer membrane. Removal of the mitochondrial-targeting domain of cardif abolishes its ability to induce IFNs. Cardif is cleaved and inactivated by NS3-4A, a serine protease from hepatitis C virus known to block interferon-β production.
