General Information
RIG-I (retinoic acid-inducible gene I; Ddx58), a member of the DEAD-box RNA helicases, is activated by cytoplasmic dsRNA produced during the viral replication. The protein is characterized by a N-terminal region with two caspase recruitment domains (CARD) and a C-terminal region harboring potential ATP-dependent RNA helicase activity. RIG-1 recruits the CARD adaptor inducing IFN-β (Cardif) in a CARD-CARD-dependent manner resulting in NF-κB and IRF3 activation.

General Information
Certain serine/threonine protein kinases, such as ASK1, RIP, DAP, and ZIP kinases, are mediators of apoptosis. Receptor interacting proteins including RIP and RIP2/RICK mediate apoptosis induced by TNF-R1 and Fas, two prototype members in the death receptor family. A novel member in the RIP kinase family was recently identified and designated RIP3 [1-3]. RIP3 contains an N-terminal kinase domain but, unlike RIP or RIP2, lacks the C-terminal death or CARD domain. RIP3 binds to RIP and TNF-R1, mediates TNF-R1 induced apoptosis, and attenuates RIP and TNF-R1 induced NF-κB activation. Overexpression of RIP3 induces apoptosis and NF-κB activation. The messenger RNA of RIP3 is expressed in a subset of adult tissues [2,3]. Meanwhile RIP4 has been identified and initially characterized. These recent data suggest that RIP4 is involved in NF-κB and JNK signalling and that caspase-dependent processing of RIP4 may negatively regulate NF-κB-dependent pro-survival or pro-inflammatory signals [4].

General Information
In mammals, TIR domain adapters, including Myd88, TIRAP, TICAM-1 (also called TRIF), and TRAM, act in Toll-like receptor (TLR) signaling pathway. The last member of this family called SARM (sterile and armadillo motifs) is highly conserved but has no known function in any organism. SARM is the only mammalian TIR-domain adapter proteins conserved in C. elegans, suggesting that SARM is the most ancient member of this class of signaling proteins. Overexpression of SARM fails to activate either NF-kB- or IRF-3-dependent reporter gene expression, two transcription factors that are sensitive to TLR signaling, suggesting that SARM is functionally distinct from other mammalian TIR domain-containing proteins.

General Information
Toll-like receptors (TLRs) play an essential role in the detection and elimination of invading pathogens. Recognition of microbial components by TLRs triggers activation of the innate immune system. Upon stimulation, these receptors interact with specific TIR domain-containing adaptor proteins. Five such adaptors have been discovered to date, MyD88, Trif (TIR-domain-containing adaptor inducing interferon-β), TIRAP (TIR domain-containing adaptor protein)/Mal (MyD88 adaptor-like), TRAM (Trif-related adaptor molecule) and SARM (SAM and ARM-containing protein). MyD88 is common to all TLR-mediated pathways, which lead to the production of inflammatory cytokines, whereas TRIF mediates induction of IFN-β in TLR3 and TLR4 signalling pathways. TIRAP/Mal is implicated in the TLR2- and TLR4-mediated MyD88-dependent signalling pathway. TRAM is specifically involved in the TLR4-mediated TRIF-dependent pathway. Thus, TIR domain-containing adaptors provide specificity of TLR signalling.
