IMMUNOGEN:	Recombinant rat adiponectin.
SPECIFICITY:	Recognizes both natural and recombinant rat adiponectin. Does not cross-react with human and mouse adiponectin. Detects a band of ~30kDa by Western blot.
APPLICATION:	ELISA (direct and indirect: 1:5’000-1:10’000). Western Blot (1:2’000-10’000 using ECL. Suggested blocking and dilution buffer is TBST containing 0.05% Tween 20 and 5% skim milk. Suggested incubation time is 1 hour at room temperature.) Optimal conditions must be determined individually for each application.

BACKGROUND/TECHNICAL INFORMATION

Western Blot Protocol:

1: SDS-PAGE Sample buffer
62.5mM Tris-HCl (pH 6.8), 2% SDS, 10% glycerol, 5% β-mercaptoethanol, 0.1% (w/v) bromophenol blue.

2: Serum preparation
A. 1µl rat serum and 9µl sample buffer after boiling for 5 min.
B. Recombinant rat adiponectin protein: 100ng/µl and 9µl sample buffer after boiling for 5 min.

3: Electrophoresis

4: Transfer
A. Nitrocellulose membrane
B. Constant 30V for 16 hours.

5: Western blotting
A. Blocking: 0.5% skim milk in TBST
B. Secondary IgG: anti-mouse IgG whole molecular (1:5’000)
C. ECL reaction
