General Information
BAG (Bcl-2-associated athanogene) family proteins contain a BAG domain and interact with a variety of partners including heat shock proteins, steroid hormone receptors, Bcl-2 and Raf-1. BAG-1 and BAG-3 participate in a wide variety of cellular processes including signalling, transcription, stress response, and apoptosis. BAG-3 (CAIR-1; Bis) forms a complex with HSC70 and assists in polypeptide folding. In addition, this complex can mediate protein delivery to proteasome and is able to downregulate apoptosis by interfering with cytochrome c release, apoptosome assembly and other events. In human cancer cell lines, BAG-3 expression sustains cell survival and underlies resistance to therapy through downmodulation of apoptosis.
