Product Description
Ski [1] and SnoN [2] are oncoproteins and transcription factors that form both homodimers and heterodimers [3]. Both have the ability to transform avian fibroblasts and induce muscle differentiation [4,5]. They recognize a specific DNA binding site (GTCTAGAC) through which they repress transcription [6]. Ski and SnoN are components of a macromolecular complex containing the co-repressor N-CoR/SMRT, mSin3 and histone deacetylase. Ski and SnoN negatively regulate TGF-β signalling by recruiting this complex to Smads [7]. Interactions between Ski and Smad2, -3 and -4 blocks Smads-mediated transcriptional activation as well as TGF-β-induced growth suppression of epithelial cells, suggesting oncogenic effects of Ski derive in part through its ability to antagonize the growth-inhibitory signals initiated by members of the TGF-β family of ligands.
