Product Specification
SOURCE/HOST: Smooth (S)-form LPS, isolated and purified from Salmonella abortus equi by modification of the phenol water extraction and PCP method [1]. LPS-Biotin was prepared using the biotin reagent biotinamidocaproate N-hydroxysuccinimide ester. Briefly, LPS at 10mg/ml in distilled water was mixed with biotin reagent in sodium bicarbonate buffer. The reaction mixture was stirred, dialyzed extensively against distilled water in the dark, and sterile filtered.
CONCENTRATION: 1mg/ml
PURITY: Absence of dectectable protein or DNA contaminants with agonistic TLR activity.
FORMULATION: Liquid. Sterile, ready-to-use solution in pyrogen-free double-distilled water.

HANDLING: After opening, prepare aliquots and store at -20°C. Use must be restricted to qualified personnel. Keep sterile. Do not ingest. Wear gloves and mask when handling this product! Avoid contact through all modes of exposure. LPS compounds are highly pyrogenic. Avoid accidental injection; extreme care should be taken when handling in conjunction with hypodermic syringes. Avoid inhalation of LPS and prevent these compounds from entering the bloodstream.

BACKGROUND/TECHNICAL INFORMATION

Note: As the biotinylation reagent and standard biotinylation protocol favors protein/peptide (contaminants) about 1000-fold over sugar as a target, it is mandatory to use >99.9% pure LPS to exclude the biotinylation of contaminants. These are usually present in many commercial LPS preparations and would lead to misleading observations of the tracking/staining with the reagents.
TLRgrade™ LPS fulfills such high quality requirements and the biotinylation procedure is chosen to preserve TLR4 (Toll-like receptor 4) binding and cellular activation using TLRgrade™ LPS-Biotin.

General Information
BACKGROUND/TECHNICAL INFORMATION Flow Cytometry Protocol:
- Brief centrifugation before use is recommended
- Wash cells in PBS by gentle shaking or pipetting up and down
- Resuspend cells in binding buffer (10mM HEPES/NaOH, pH 7.4, 140mM NaCl, 2.5mM CaCl2; filtered
through 0.2µm pore filter); adjust cell density to 2-5x105/ml
- Take 195µl cell suspension and add 5µl Annexin V-Biotin
- Mix and incubate 15 minutes at room temperature
- Wash cells 2x in binding buffer
- Resuspend in binding buffer
- Add Streptavidin-FITC
- Mix and incubate for 30 minutes in the dark at room temperature
- Wash cells 1x in binding buffer and resuspend in 190µl binding buffer
- Add 10µl of 20µg/ml propidium iodide stock solution (end concentration 1mg/ml)
- FACS analysis
