General Information
Caspase-1 is the prototypical member of the caspase family of cysteine proteases. Caspase-1 exists in cells as an inactive 45kDa pro-enzyme. The pro-enzyme is matured by proteolysis to yield large (20kDa) and small (10kDa) subunits. The active enzyme is a heterotetramer consisting of two large and two small subunits. To date the mechanism of regulation of caspase-1 activation is complex and poorly understood. In THP-1 cells, a large proportion of the caspase-1 is present in the inactive pro-enzyme form.

General Information
Caspase-2 is a member of the interleukin-1β converting enzyme (ICE) family of cysteine proteases. Similar as other caspases, caspase-2 also exists in cells as an inactive proenzyme. During apoptosis, procaspase-2 is processed at aspartate residues by self-proteolysis and/or cleavage by upstream caspases. The processed form of caspase-2 consists of large (19kDa) and small (12kDa) subunits which associate to form the active enzyme. The expressed caspase-2 spontaneously undergoes auto-processing to yield the subunits characteristic of the native enzyme. The active recombinant caspase-2 is routinely tested for its ability to enzymatically cleave the caspase-2 colorimetric substrate Ac-VDVAD-pNA (Prod. No.

). The signal can be detected by spectrophotometry.

Enzyme Evaluation Protocol
This protocol is used to measure caspase enzyme activity. The synthetic fluorogenic peptide Ac-VDVAD-AFC (Prod. No.

) is used as the caspase enzyme substrate. The enzyme cleaves the substrate releasing the fluorescent AFC (7-amino-4-trifluoromethylcoumarin). AFC release can be measured by spectrofluorometry using UV.
1. Add 20mM of Ac-VDVAD-AFC (Prod. No.

) to 1ml of assay buffer (20mM PIPES, 100mM sodium chloride, 10mM DTT, 1mM EDTA, 0.1% (w/v) CHAPS, 10% sucrose, pH 7.2). Add the appropriate amount of active caspase to the mixture (50ng/ml).
2. Incubate for 1 hour at 37°C.
3. Measure the AFC liberated from the Ac-VDVAD-AFC using a spectrofluorometer with an excitation wavelength of 400nm and an emission wavelength of 505nm.

General Information
Caspase-3 is a member of the interleukin-1β converting enzyme (ICE) family of cysteine proteases. It exists in cells as an inactive 32kDa proenzyme. During apoptosis procaspase-3 is processed at aspartate residues by self-proteolysis and/or cleavage by upstream caspases, such as caspase-6, caspase-8 and granzyme B. The processed form of caspase-3 consists of large (17kDa) and small (11kDa) subunits which associate to form the active enzyme. Active caspase-3 has been shown involved in the proteolysis of several important molecules, such as poly(ADP-ribose) polymerase (PARP), the sterol regulatory element binding proteins (SREBPs), focal adhesion kinase (FAK) and others.
