APPLICATION: Useful in screening caspase inhibitors, studying enzyme regulation and kinetics, determining target substrate or as a positive control in caspase activity assays. We recommend using 1 unit per assay for analyzing caspase activity.

General Information
Caspase-4 is a member of the caspase family of cysteine proteases. It exists in cells as an inactive pro-enzyme. The pro-enzyme is matured by proteolysis to yield large and small subunits. The active enzyme is a heterotetramer consisting of two large and two small subunits. The expressed caspase-4 spontaneously undergoes auto-processing to yield subunits characteristic of the native enzyme. The active caspase-4 is routinely tested for its ability to enzymatically cleave the substrate WEHD-AFC or WEHD-pNA.

General Information
BAP31 (B cell receptor-associated protein 31) is an integral protein of the ER membrane that forms a large hetero-oligomeric complex with the related BAP29. The cytosolic domain of BAP31 contains two identical caspase recognition sites that are preferentially cleaved by initiator caspases, including caspase-8. After activation of cell surface death receptors, human BAP31 is cleaved into a membrane-embedded fragment called p20, which induces apoptosis when expressed ectopically. The p20 fragment can mediate Ca2+-dependent apoptotic cross-talk between the ER and mitochondria, stimulating cytochrome c release. In addition to its role in apoptosis, BAP31 is a component of the ER quality control compartment and functions as a cargo receptor for ER export of transmembrane proteins such as cellubrevin, MHC class I molecules, and cytochrome P450 2C2.

Calnexin, also referred to as IP90 or p88, is a ~88kDa chaperone protein found in the endoplastmic reticulum. It binds with partially folded immunoglobulin proteins and retains them in the endoplastmic reticulum until they are completely folded.

SPECIFICITY: Recognizes an epitope within the p20 fragment of mouse caspase-12. Detects a band of ~51kDa by Western blot. Does not cross-react with mouse caspase-1 or -11.
