Abstract
General Information
BAP31 (B cell receptor-associated protein 31) is an integral protein of the ER membrane that forms a large hetero-oligomeric complex with the related BAP29. The cytosolic domain of BAP31 contains two identical caspase recognition sites that are preferentially cleaved by initiator caspases, including caspase-8. After activation of cell surface death receptors, human BAP31 is cleaved into a membrane-embedded fragment called p20, which induces apoptosis when expressed ectopically. The p20 fragment can mediate Ca
2+
-dependent apoptotic cross-talk between the ER and mitochondria, stimulating cytochrome c release. In addition to its role in apoptosis, BAP31 is a component of the ER quality control compartment and functions as a cargo receptor for ER export of transmembrane proteins such as cellubrevin, MHC class I molecules, and cytochrome P450 2C2.

General Information
Caspase-12 is a member of the ICE/CED-3 family of caspases and is detected as a protein of ~51kDa. Overexpression of caspase-12 in cells induces apoptosis. Capase-12 is localised to the endoplasmatic reticulum (ER) and activated by ER stress and specifically cleaves and activates pro-caspase-9 and is also thought to be involved in the apoptosis induced by viral infection. Caspase-12-deficient mice are resistant to ER stress-induced apoptosis. Caspase-12 is probably expressed in many and perhaps all mouse tissues; and is expressed at high levels in skeletal muscle, heart, lung and kidney, and at moderate levels in brain and liver. In many humans caspase-12 is a pseudo gene and not expressed.

General Information
CHOP is a small nuclear protein that is capable of dimerizing with transcription factors C/EBP α and β. Once dimerized, this complex inhibits the normal binding and function of C/EBP to classical binding sites. Inversely, the C/EBP-CHOP dimer gains binding activity to other non-classical C/EBP stress related targets.
Under normal cellular conditions this protein is not expressed in detectable levels, but is highly unregulated during times of cellular/ER stress. Examples of CHOP inducing stress include: treatment with tunicamycin, nutrient starvation and reducing agents that interfere with the calcium flux across the ER membrane.
