General Information
The Ets family of transcription factors is known to control the expression of genes critical for cellular proliferation, differentiation, and transformation. They activate transcription by interaction with purine-rich sequences in promoter/enhancer regions of various genes. Differential expression of Ets genes is particularly associated with embryonic development of the central nervous system. Previous studies have implicated Ets transcription factors in apoptosis; however, exact mechanisms remain unclear. The mammalian Ets1 gene encodes a set of nuclear phosphoproteins ranging in size from 42 to 54kD.

General Information
Myc proteins are important regulators of cell behaviour and work as transcription factors that belong to the basic region/helix-loop-helix/leucine zipper family of DNA binding proteins. To regulate its target genes, Myc recruits several different cofactors (e.g. PARP-10). They control a variety of cellular functions including protein synthesis, cell cycle progression, and apoptosis, thereby modulating cell growth and proliferation, differentiation, and survivial of cells. The family of myc genes includes B-myc, L-myc, N-myc and s-myc; however only c-myc, L-myc and N-myc have neoplastic potential.
The c-Myc gene was discovered as the cellular homolog of the retroviral v-myc oncogene. It is a proto-oncogene, whereas the deregulated expression of c-Myc is observed in the majority of human and animal tumors often with poor prognosis.
The proto-oncogene c-Myc is implicated in various physiological processes; cell growth, proliferation, loss of differentiation and cell death (apoptosis). It also has been implicated in the loss and dysfunction of insulin-producing beta cells in diabetes. Recent studies in mice suggest that c-Myc has unexpected functions during both self-renewal and the differentiation of stem and early progenitor cells. c-Myc activity is normally tightly controlled by external signals including growth factors, mitogens and beta-catenin which promote, and factors as TGF-beta which inhibit.

LEF is a transcription factor of the LEF/TCF protein family. LEF interacts with cytoplasmic β-catenin and regulates the expression of genes containing a LEF/TCF binding site. The formation of LEF-β-catenin heterodimers is an important step in the wnt/wg signal transduction pathway.
