Product Description
Nitrotyrosine is the major product from the spontaneous reaction of peroxynitrite with tyrosine. Formation of nitrotyrosine in vivo can indicate the formation of peroxynitrite by a nitric oxide (NO)-dependent oxidative damage.

BACKGROUND/TECHNICAL INFORMATION Amount of protein loaded may need to be adjusted depending on the antibody concentration, the detection method utilized (i.e. ECL or color precipitation), and the amount of time required to visualize nitrotyrosine from experimental samples on the film or membrane. E. g., at an antibody concentration of 1mg/ml, 50 ng of peroxynitrite-treated BSA forms a dark band using the NBT/BCIP detection method.

General Information
Nitric oxide (NO) is a product of the enzymatic conversion of arginine to citrulline by nitric oxide synthase. NO reacts rapidly with superoxide (6.7 x 109/M/sec) to form peroxynitrite. At physiological pH and in the presence of transition metals, peroxynitrite undergoes heterolytic cleavage to form hydroxyl anion and nitronium ion, the latter of which nitrates protein tyrosine residues. Thus, the presence of nitrotyrosine on proteins can be used as a marker for peroxynitrite formation in vivo. Nitrotyrosine has been shown to be present in proteins from a variety of clinical conditions including atherosclerotic lesions of human coronary arteries, postischemic heart, and placenta during preeclampsia. Increased nitration of proteins in motor neurons has been identified in patients with ALS (amyotrophic lateral sclerosis) and may be due to mutations in superoxide dismutase.

General Information
BACKGROUND/TECHNICAL INFORMATION Typical procedure for Immunoprecipitation:

1. Dilute the protein sample (cell lysate, tissues homogenate, plasma, etc.) to ~1mg/ml in TBS or PBS in a
     microcentrifuge tube.
2. Add 20-40µl of gel slurry to 0.5-1ml (0.5-1mg protein) and incubate 1-2 hours at room temperature or
     4°C overnight with occasional mixing or gentle rocking.
3. Collect the beads by brief centrifugation (<1 minute) in a microcentrifuge. Carefully drain or pipet off the
     supernatant and wash the beads 3 times with TBS or PBS.
4. Resuspend the beads in 25-50µl of 1X Laemmli sample buffer and boil for 5 minutes. (Alternatively the
     samples can be frozen for later use).
5. Briefly centrifuge the sample to pellet the beads. The supernatant can now be directly used for
     SDS-PAGE.
