APC-labelled recombinant human annexin V is suitable for dual staining experiments to detect phosphatidylserine whilst cell-surface protein can be detected with a suitable MAb (FITC-labelled or labelled secondary antibody). The fluorochrome APC (allophycocyanin) shows far red fluorescence (Ex(max): 650nm; Em(max): 660nm).

BACKGROUND/TECHNICAL INFORMATION
Flow Cytometry Protocol:
- Prepare binding buffer (10mM Hepes/NaOH, pH 7.4, 140mM NaCl, 2.5mM CaCl
2
;
filtered through 0.2µm pore filter);
- Brief centrifugation before use is recommended
- Wash cells in PBS by gentle shaking or pipetting up and down
- resuspend cells in binding buffer; adjust cell density to 2-5x10

- Take 195µl cell suspension and add 5µl Annexin V-FITC
- Mix and incubate 10 min. in the dark at room temperature
- Wash cells 1x with PBS and resuspend in 190µl binding buffer
- FACS analysis

Flow Cytometry Protocol:
- Wash cells in PBS, and resuspend in binding buffer (10mM HEPES/NaOH, pH 7.4, 140mM NaCl, 2.5mM CaCl
2
; filtered through 0.2µm filter).
- Adjust cells to a density between 200’000 and 500’000 cells/ml.
- Take 195µl cell suspension and add 5µl Annexin-ATTO 488. Mix and incubate 10 min. in the dark.
- Wash cells 1x and resuspend in 190µl binding buffer.
- Add 10µl of 20µg/ml propidium iodide (PI) stock solution to stain for necrotic cells.
- Analyze by FACS.

- Wash cells 2x in binding buffer

- Add Streptavidin-FITC

- Wash cells 1x in binding buffer and resuspend in 190µl binding buffer
- Add 10µl of 20µg/ml propidium iodide stock solution (end concentration 1mg/ml)
- FACS analysis
