Note:
The use of DMSO may abolish the iGb3 activity and should be avoided [2].
For expanding human NKT cells from freshly isolated peripheral blood mononuclear cells (PBMC)
:
Mix iGb3 at 100ng/ml with PBMC and culture for 4-5 days.
For the stimulation of mouse NKT cells
:
The bone marrow derived dendritic cells (BMDC) is the most efficient cell type that present iGb3. Use 50’000-250’000 of BMDCs pulsed overnight with 100ng/ml of iGb3 and 50’000 freshly purified NKT cells as a responder. Use tetramer sorting to purifiy the NKT cells from spleen and before mixing with BMDCs, rest them in culture medium for 12 hours and then wash with medium to avoid a high background. MACS sorting technique is not recommended. Alternatively, NKT cell hybridomas can be used to detect the ligand. (For reference see:

Naturally occurring ceramide and an integral part of the sphingomyelin cycle [1]. C

-N-acyl sphingosines are the most abundant ceramide species in human plasma [2] and in rabbit leukocytes [3], where they make up to 50-60% of total ceramides. Lignoceric ceramide stimulates the proliferation of basophilic erythroblasts, suggesting a possible role in erythrocyte maturation [3].

Abstract
General Information
Natural killer T cells (NKT cells) are important regulators of the immune system. They share properties of both innate as well as adaptive immunity [1]. They express T cell receptors (TCR) on their surface consisting of an invariable α-chain and a somewhat variable β-chain [2]. The majority of NKT cells express the invariable α-chain Vα14-Jα18 (Vα14) in mouse or its homolog in humans Vα24-Jα18 (Vα24) [3]. Vα14 NKT cells recognize α-GalCer presented by CD1d [4, 5].
Recently, NKT cells expressing the invariant Vα19-Jα26 TCR α-chain (Vα19 NKT cells) have been found in peripheral blood lymphocytes and in the liver [6]. They recognize α-ManCer presented by MR1 (Major histocompatibility complex, class I-related) and respond with production of large amounts of immunoregulatory cytokines [7, 8].
