General Information
The adaptor protein ARAP1 is a PIP3-dependent Arf GAP that regulates Arf-, Rho-, and Cdc42-dependent cell activities. ARAP1 plays an important role in the regulation of blood pressure and renal function via activation of the intrarenal renin-angiotensin system. It was shown that ARAP1 might affect the efficacy of TRAIL-induced apoptosis due to its cell-specific trafficking of TRAIL-R1 (DR4).

Product Description
RasGAP
RasGAP (Ras-specific GTPase-activating protein) is one of the few signalling proteins cleaved by caspases during apoptosis [1-3]. The kinetics of RasGAP cleavage is identical to well-known caspase substrates such as PARP or the caspase themselves. RasGAP cleavage by caspases generates a carboxy-terminal fragment of about 64 kDa (fragment C) containing the GAP activity and a N-terminal fragment (fragment N) of about 56 kDa. The latter fragment can be further cleaved into two other smaller fragments, one of which contains the SH2 and SH3 domains. The fact that RasGAP is an abundant ubiquitous protein readily cleaved during apoptosis makes it a very good apoptotic marker.
RasGAP is a unique caspase substrate because it can either inhibit or promote apoptosis depending on the extent of its cleavage by caspases. At low levels of caspase activity, generation of fragment N induces a potent Ras-PI3K-Akt-dependent survival pathway. At higher levels of caspase activity, fragment N is further cleaved into two fragments (N1 and N2) that potentiate apoptosis. RasGAP has recently been qualified as an “apoptostat” because it functions as a sensor of caspase activity to determine whether or not a cell should survive [2].

General Information
BACKGROUND/TECHNICAL INFORMATION Technical note: Due to the conformation of native full-length RasGAP, steric hinderance prevents binding of this antibody to the C-terminus. Thus immunoprecipitation is possible only with the C-terminal RasGAP fragment and not with the full-length protein.

The C-terminal (aa 456-1047) fragment contains the GTPase-activating protein activity of RasGAP as well as a C2 domain and a PH domain.

Product Description
SynGAP, synaptic Ras-GTPase activating protein, is a PDZ-domain interacting protein expressed almost exclusively in the brain. SynGAP has been shown to be a major component of postsynaptic density and associates with PSD-95, a synaptic scaffold protein, and synaptic NMDA receptors. SynGAP Ras-GTPase activity has also been shown to be inhibited by phosphorylation by CaM kinase II, another abundant protein located in the postsynaptic density. It has been proposed that SynGAP may function as a negative regulator of MAP Kinase activation in the absence of active NMDA receptors.
