General Information
The receptors of the UNC family (for uncoordinated) include UNC-5 of C. elegans and its four vertebrate homologs, UNC5A, B, C and D (previously called UNC5H1-4). The UNC5 family binds to the netrin family of ligands and mediates axon repulsion effect of netrin, either alone or by binding to the DCC (another netrin receptor). These receptors are type I transmembrane proteins of roughly 110-120kDa composed of two extracellular Ig-like domains, two thrombospondin type I domains, and intracellular sequence with a ZU5 domain and a death domain. UNC5B is a direct target gene for p53 that mediates p53-dependent apoptosis, and is also a dependence receptor that regulates apoptosis either positively or negatively depending on its binding with netrin-1. UNC5B is highly expressed in brain, but also expressed at lower level in developing lung, cartilage, kidney, hematopoietic and immune tissues, as well as in the vascular system. UNC5B (as well as the other UNC5 receptors) is a putative tumour suppressor because of its frequent alteration and reduced expression in various human cancers. Recently UNC5B has been shown to mediate guidance events controlling the morphogenesis of the vascular system.
