General Information
An aspartyl protease has been identified as the β-secretase of the amyloid precursor protein (APP), cleavage of which at the appropriate site leads to the deposition of amyloid plaques found in the neurodegenerative and dementia-inducing disorder known as Alzheimer's disease. Endoproteolysis of APP leads to the production of a 4kDa, insoluble polypeptide known as amyloid β-peptide (Aβ). Aβ is the major constituent of the senile plaques and intracellular neurofibrillary tangles seen in the cerebral cortex and hippocampus of affected individuals. β-Secretase, which has also been termed beta-site APP-cleaving enzyme (BACE), has been shown to co-localize with the Golgi markers β-Coat Protein (β-COP) and furin convertase, and with the endosomal resident protein EA1 which is consistent with its participation in the processing of the secreted Aβ polypeptide. This protein is also glycosylated and possesses a transmembrane domain which is C-terminal to the active site found within the lumen of the Golgi and endosomes.
