General Information

RANKL (receptor activator of nuclear factor (NF)-κB ligand; also: osteoprotegerin ligand, OPGL), its cellular receptor, receptor activator of NF-κB (RANK), and the decoy receptor, osteoprotege-rin (OPG) have been identified as the key molecular regulation system for bone remodelling. RANKL, a member of the tumor necrosis factor (TNF) family, is the main stimulatory factor for the formation of mature osteoclasts and is essential for their survival. Therefore, an increase in RANKL expression leads to bone resorption and bone loss. RANKL is produced by osteoblastic lineage cells and activated T lymphocytes. It activates its specific receptor RANK which is located on osteoclasts and dendritic cells.
The effects of RANKL are counteracted by OPG which is secreted by various tissues and acts as an endogenous soluble receptor antagonist.
Imbalances of the RANKL/OPG system have been related to the pathogenesis of Paget’s disease, benign and malignant bone tumors, postmenopausal osteoporosis, rheumatoid arthritis, bone metastases and hypercalcemia. It was shown in several studies that in animal models restoring of the RANKL/OPG balance (e.g. by administering OPG) reduces the severity of these disorders.
It has been shown, that RANKL is produced as a membrane-bound protein on murine osteoblasts/stromal cells, and cleaved into a soluble form by a metalloprotease. Stimulators of the osteoclastogenesis such as IL-1β, IL-6, IL-11, IL-17, and TNF-α, increase the expression of RANKL and decrease OPG expression in osteoblasts/stromal cells. Cytokines inhibiting the osteoclastogenesis such as IL-13, INF-γ, and TGF-β1, suppress the expression of RANKL and stimulated OPG expression.

Molecular structure:
sRANKL is a part of the TNF superfamily with high similarity to other members of that protein species. (SwissProt Nr. O14788). Two isoforms are produced by alternate splicing, a type II mem-brane protein (ISOFORM 1, 317 AA, MW 35.5 kD), and a secreted molecule (ISOFORM 2, 244 AA, MW 27.7kD), lacking the cytoplasmic and transmembrane domain. Although both forms are bioactive, the membrane bound protein seems to be the homeostatic form, while the produc-tion of soluble RANKL signals pathological conditions.

General Information
BAFF (B cell activation factor of the TNF family, also known as BLyS or TALL1) is a cytokine expressed predominantly by cells of the immune system such as neutrophils, monocytes, macrophages, dendritic cells, follicular dendritic cells, activated T cells and some malignant B cells. General Information
TL1A (TNFSF15), a ligand belonging to the tumor necrosis factor (TNF) family, is expressed predominantly by endothelial cells and monocytes [1]. TL1a is upregulated by the proinflammatory cytokines TNF and IL-1 and also by immune complexes (IC) [2]. TL1 functions in T cell costimulation and Th1 polarization. On activated T cells, TL1A functions specifically via its surface-bound receptor DR3, (a member of the death-domain containing TNF receptor family) to promote cell survival and secretion of proinflammatory cytokines (1). The secreted decoy receptor 3 (DcR3), a soluble protein of the tumor necrosis factor receptor (TNFR) superfamily blocks the action of TL1A [3]. TL1A can be released to circulate as a homotrimeric soluble form [4]. Activation of DR3 by TL1A induced the formation of a signaling complex containing TRADD, TRAF2, and RIP and activated the NF-κB and the ERK, JNK, and p38 mitogen-activated protein kinase pathways [5]. The TL1A/DR3 pathway plays an important role in Th1-mediated intestinal diseases, such as Crohn’s disease [6].

General Information
OPG (Osteoprotegerin; also called Osteoclastogenesis Inhibitory Factor or TNFRSF11B) is a secreted member of the TNF (Tumor necrosis factor) receptor superfamily of proteins [1]. OPG, composed of 380 amino acids, is expressed as monomer and secreted as disulphide-homodimeric protein [2]. Produced by numerous cell types and tissues, its expression pattern targets the skeletal, immune, and vascular systems. Osteoblasts secrete OPG that protects the skeleton from excessive bone resorption by acting as a decoy receptor for the ligand nuclear factor-κB ligand (RANKL), preventing it from interacting with its receptor RANK [3]. OPG/RANK/RANKL molecular triumvirat plays a pivotal function in the regulation of the immune system through dendritic cells [4]. OPG-deficient mice exhibit severe osteoporosis and vascular calcification of the aorta and renal arteries [5]. Defects in TNFRSF11B are the cause of juvenile Paget disease (JPD), a bone disease [6]. OPG also functions as a soluble receptor for the TNF ligand TRAIL (TNF-related apoptosis-inducing ligand) preventing it to activate apoptotic signaling cascase and therefore providing cancer cells with a survival advantage [7].

General Information
LIGHT (derived from "homologous to Lymphotoxins, exhibits Inducible expression, and competes with Herpes simplex virus (HSV) Glycoprotein D for Herpes virus entry mediator (HVEM /TR2), a receptor expressed by T lymphocytes")is a 29 kDa type 2 transmembrane protein that belongs to the TNF superfamily [1, 2]. It is expressed in granulocytes, monocytes, platelets, immature dendritic cells and activated T cells. LIGHT is active either as membrane-anchored or secreted (soluble) forms. Three receptors of the TNF superfamily: lymphotoxin receptor (LTR), herpesvirus entry mediator (HVEM), and decoy receptor 3 (DcR3) binds to the ligand. LIGHT is involved in T-cell regulation and dendritic cell maturation [3]. It triggers several signaling pathways such as NF-κB, non-canonical NF-κB, Stat3, ERK5 / BMK1 and JNK [4]. LIGHT is released from platelets and induces a proinflammatory state in vascular endothelial cells [5]. It suppress tumor formation in vivo by inducing apoptotic cell death. Embryonic stem (ES) cells are differentiated in the presence of LIGHT [6]. Mice overexpressing LIGHT spontaneously develop severe autoimmune disease. LIGHT has been reported to increase in Rhumatoid Arthritis (RA) patients and to induce a RANKL ­dependent and -independent osteoclasts formation [7].

Several studies have reported that visceral obesity (fat located within omental and mesenteric fat depots) is more closely associated with insulin resitance, type 2 diabetes and cardiovascular disease than is peripheral obesity (subcutaneous) [2]. However, the underlying mechanism is still unclear. Recently, a new protein Omentin 1 (also named Omentin, Intelectin-1, Endothelial Lectin HL-1 and Intestinal Lactoferrin Receptor) has been identified as a major visceral (omental) fat secretory adipokine [3]. Omentin 1 is a secreted protein of ~38kDa containing a fibrinogen-related domain that is highly expressed in the omental fat, and much less in intestine, lung and heart. Addition of recombinant Omentin 1 in vitro does not affect basal, but enhances insulin-stimulated glucose uptake in subcutaneous as well as in omental human adipocytes. Omentin 1 triggers AKT signalling in the absence of insulin [3]. Omentin 1 plasma levels measured by western blotting are decreased in obesity [4]. Omentin 1 is decreased in patients with polycystic ovary syndrome (PCOS), a disease associated with insulin resistance and obesity [5]. Glucose and Insulin negatively regulate Omentin-1 levels ex vivo and in vivo [5].
