Product Description
Inhibits translocation of the NF-κB active complex into the nucleus. In mouse endothelial LE-II cells induced with LPS, inhibition of NF-κB nuclear translocation was 88% at 50µg/ml (18µM). In human monocytic THP-1 cells induced by LPS or TNF-α there was 85% inhibition at 100µg/ml. For control peptide, see SN50M (Prod. No. ALX-167-025).

Inactive control peptide for SN50 (Prod. No. ALX-167-024). In mouse endothelial LE-II cells, induced with LPS, 50µg/ml SN50M had no measurable effect on NF-κB translocation. In human monocytic THP-1 cells induced by LPS or TNF-α there was slight inhibition at 100µg/ml.

General Information
Caspase-1 is the prototypical member of the caspase family of cysteine proteases. Caspase-1 exists in cells as an inactive 45kDa pro-enzyme. The pro-enzyme is matured by proteolysis to yield large (20kDa) and small (10kDa) subunits. The active enzyme is a heterotetramer consisting of two large and two small subunits. To date the mechanism of regulation of caspase-1 activation is complex and poorly understood. In THP-1 cells, a large proportion of the caspase-1 is present in the inactive pro-enzyme form.

General Information
Early Growth Response- (EGR-) proteins represent a family of related transcription factors with almost identical DNA- binding zinc finger domains, but with different flanking regions. All four human EGR-proteins, EGR-1 to EGR-4 bind the EGR-consensus sequence GCG T/GGG GCG. However individual EGR-proteins show different binding to related sequences, such as the overlapping EGR-1/Sp1 binding site in the human IL-2 gene promoter. In functional assays the EGR-4 protein shows superior performance for Electrophoretic Mobility Shift Assay (EMSA) compared to proteins expressed in E.coli.
BACKGROUND/TECHNICAL INFORMATION For EMSA labeled oligonucleotides are required, that include the specific target sequence. Radioactive labeling of the oligonucleotide is performed by kinase reaction (T4 kinase) using either 32P-γ ATP or 33P-γ ATP as a source for radioactivity. Due to the lower radiation load we recommend the use of 33P-γ ATP, which results in specific, detectable bands. Formation of a specific protein: DNA complex results in a lower mobility of this complex compared to the mobility of the free DNA. The specificity of the EMSA can also be analyzed by competition experiments in which an excess of unlabeled specific oligonucleotide interferes with the formation of a specific band, while the same amount of unrelated or mutated oligonucleotide has no effect. DNA-Protein complexes are then separated from the free (unbound) oligonucleotides by native gel-electrophoresis.

RECONSTITUTION: Reconstitute to 1U/µl with PBS containing 15% glycerol.
SPECIFIC ACTIVITY: ~5'000U/mg protein. One unit is defined as the amount of enzyme that cleaves 1nmol of the caspase substrate WEHD-pNA per hour at 37°C in a reaction solution containing 50mM HEPES, pH 7.2, 50mM NaCl, 0.1% CHAPS, 10mM EDTA, 5% glycerol and 10mM DTT.
APPLICATION: In combination with caspase-5 activity assays, the enzyme is useful in screening caspase inhibitors, studying enzyme regulation and kinetics, determining target substrate or as a positive control in caspase-5 activity assays. We recommend using 1 unit per assay for analyzing caspase activity.
