Advantages: One-step, ready-to-use solution, no radioisotopes, no solubilization step, high sensitivity, correlates with the [³H]-thymidine incorporation assay. Product Description

Kit utilizes Live-dye™, a cell permeable green fluorescent dye (Ex(max): 488nm; Em(max): 518nm), to stain live cells. Dead cells can be easily stained by propidium iodide (PI), a red fluorescent dye (Ex(max): 488nm; Em(max): 615nm), which in viable cells is actively pumped out of the cytoplasm. Stained cells can be visualized by fluorescence microscopy using a band-pass filter (detects FITC and rhodamine). Cyto-dye has also been used to discriminate between apoptotic and non-apoptotic cells. Ready-to-use.

Product Description

Mammalian 26S proteasome purified through a series of specific chromatographic steps yields a partially purified fraction containing 26S proteasome and no conjugation enzymes (E1, E2, E3). This stable fraction is suitable for the degradation of preformed ubiquitin-protein conjugates by the 26S proteasome degradation kit since highly purified 26S is costly and more labile.

Product Description

The enzyme fractions supplied represent the full compliment of purified conjugation enzymes (E1, E2s, and E3s) that are found in mammalian fraction II. Conjugation Fraction A contains predominantly E1 and E2 enzymes, while Conjugation Fraction B contains predominantly E3 and deubiquitinating enzymes. The enzymes have been tested and shown to work with typical [¹²⁵I]-labeled substrate proteins such as lysozyme and β-lactoglobulin. This conjugation fraction also contains ubiquitin C-terminal hydrolases (UCH) and therefore the addition of ubiquitin aldehyde (Prod. No. ALX-200-603) is recommended for the inhibition of UCHs to improve overall conjugate yield. The supplied fractions do not contain 20S or 26S proteasomes or any other protein degradation activity. If the substrate being conjugated requires E2 or E3 enzymes not found in Fraction II, the reaction can be supplemented with enzymes from Fraction I.
