Product Description
One-step flow cytometry kit for the qualitative and quantitative measurement of apoptosis in mammalian cells. One of the most easily measured features of apoptotic cells is the break-up of the genomic DNA by cellular nucleases. The large number of DNA fragments appearing in apoptotic cells results in a multitude of 3'-hydroxyl termini of DNA ends which can be identified with fluorescent tagged deoxyuridine triphosphate nucleotides (F-dUTP). The enzyme terminal deoxynucleotidyl transferase (TdT) catalyzes a template independent addition of deoxyribonucleoside triphosphates (e.g. F-dUTP) to the 3'h-hydroxyl ends of double- or single-stranded DNA. A substantial number of these sites are available in apoptotic cells providing the basis for the single step fluorescent labelling and flow cytometric method utilized in the Apo-DIRECT™ Kit.

Product Description
This reagent (MAb F7-26) allows the important discrimination between apoptosis and necrosis [1-3,6,7]. In this respect it can be regarded as a unique research tool superior to TUNEL staining [4,5]. The specificity of the assay is based upon the high sensitivity of DNA to thermal denaturation in condensed chromatin of apoptotic cells.

Although there is no change of the original product there have been important improvements in the staining protocol involving a formamide-based DNA-denaturation [8]. This is now more convenient to use, yielding a sharper contrast between apoptotic cells and controls. The increased sensitivity of MAb F7-26 allows omission of a previously recommended Biotin/Streptavidin amplification step, a source of background problems. The APOSTAIN assay is now applicable for cold (4°C) formalin fixed tissue. For recent applications of MAb F7-26 see [9-15].

Product Description
The Quick Apoptotic DNA Ladder Detection Kit offers fast and sensitive detection of DNA fragmentation in apoptotic cells. Unlike other similar kits on the market which require 1-2 days to prepare nucleic acids, the new procedure requires less than 90 minutes to prepare chromosomal DNA in a single tube, with no need for extraction or a column. DNA ladders can easily be visualized by agarose gel electrophoresis. The new procedure increases recovery of small fragmented DNA, and therefore improves detection sensitivity.
