General Information
p300 is a transcriptional coactivator closely related to CREB binding protein (CBP), interacting with many different DNA-binding transcription factors including nuclear hormone receptors, CREB (cyclic AMP-responsive enhancer binding protein), cFos, c-Jun/v-Jun, c-Myb/v-Myb, TFIIB, and MyoD. p300 displays histone acetyltransferase (HAT) activity, capable of acetylating all four core histone particles in nucleosomes.

In mammals, several genes that encode members of the basic helix-loop helix (bHLH) PAS (PER-ARNT-SIM) transcription factor family have been shown to play a significant role in regulating circadian oscillations. Transactivation of CLOCK-induced genes is mediated via an E box enhancer (CACGTG) found upstream of target genes. CLOCK-BMAL1 heterodimers bind to E box regulatory elements and stimulate gene transcription. CLOCK has been shown to transactivate the mammalian homolog of Drosophila per. PER, in concert with the product of the mammalian timeless gene (TIM), negatively regulates its own transcription by blocking the activity of the CLOCK-BMAL1 transactivation complex.

Product Description
Steroid and thyroid hormones and retinoic acid regulate a complex array of gene expression activity via intracellular receptor transcription factors belonging to the ligand dependent nuclear receptor superfamily. Adding to the complexity of function of these transcription factors are associated proteins known as coactivators and corepressors which, as their names suggest, enhance or depress transcription activity of the nuclear receptor with which they associate. Receptor-Interacting Protein 140, also known as RIP140, has been shown to enhance estrogen receptor transcriptional activity through the AF2-AD domain in an agonist dependent manner and has also been implicated to be involved in other steroid receptor pathways, including the retinoid pathway. RIP140 has recently been shown to interact with peroxisome proliferator-activated receptor-α (PPARα) and has been proposed to competitively inhibit PPARα interaction with another known nuclear receptor coactivator, SRC-1. However, the specific roles for coactivation and corepression by RIP140 in steroid receptor pathways is currently not well understood.
