General Information
BAFF (B cell activation factor of the TNF family, also known as BLyS or TALL1) is a cytokine expressed predominantly by cells of the immune system such as neutrophils, monocytes, macrophages, dendritic cells, follicular dendritic cells, activated T cells and some malignant B cells. BAFF binds three distinct receptors (BAFF-R, TACI and BCMA) expressed predominantly on B cells, although activated T cells also express BAFF-R. BAFF is a master regulator of peripheral B cell survival, and also acts in processes such as immunoglobulin isotype switch and B cell co-stimulation. Besides its major role in B cell biology, BAFF co-stimulates activated T cells. Deregulated expression of this membrane-bound protein, which can readily be released in a soluble form, leads to autoimmune disorders in mice. In humans, elevated levels of soluble BAFF have been detected in the serum of patients with various autoimmune diseases.

General Information
Chemerin (tazarotene-induced gene 2; TIG2) is a chemotactic agent that was identified as the ligand for the orphan G protein-coupled receptor ChemR23. Recently, chemerin has been shown to be expressed abundantly in adipose tissue and to function as a new adipokine [1, 2]. Chemerin potentiates insulin-stimulated glucose uptake and regulates adipogenesis and adipocyte metabolism [1].

General Information
Adipose tissue functions as storage of energy and plays an active role in monitoring and controlling whole-body metabolism by secreting a number of proinflammatory adipokines, (such as leptin, resistin, tumor necrosis factor-α and interleukin-6), as well as one anti-inflammatory adipokine, adiponectin. Adiponectin is involved in controlling the whole-body metabolism, particularly by increasing insulin sensitivity in muscle and liver, and by increasing fatty acid oxidation in muscle [1]. Recently, a new highly conserved family of adiponectin paralogs designated as C1q/Tumor Necrosis Factor-α-Related Proteins (CTRPs) 1-7, has been described in mouse and human [2-5]. This family of proteins exhibits similar structural and biological properties to adiponectin. The CTRPs, unlike adiponectin whose expression is restricted to adipose tissue, are widely expressed by many different adult tissues. Like adiponectin, CTRP2 rapidly induces acetyl-CoA carboxylase, and mitogen-activated protein kinase phosphorylation in C2C12 myotubes, which resulted in increased glycogen accumulation and fatty acid oxidation, and CTRP7 (as well as CTRPs 1, 2 and 3) induces AMP-activated protein kinase phosphorylation. The activity of the CTRPs suggests that some members of the CTRP family of proteins could substitute for adiponectin function

General Information
Interleukin-1 (IL-1) family members, such as IL-1α/β and IL-18 play important roles in host defense, immune regulation, neuronal injury and inflammation [1, 2]. Recently, a new member of this family, IL-33, also named Nuclear Factor from High Endothelial Venules (NF-HEV) [3], has been reported to be the specific ligand for ST2 [4], an orphan receptor of the IL-1 receptor family [5]. Structurally, IL-33 is closest to IL-18, rather than IL-1β. The human IL-33 is expressed with a prodomain (aa 1-111) that is cleaved by caspase-1 to produce a 18kDa mature protein (112-270) [6]. The stimulation of ST2 by IL-33 induces the NF-κB and MAPK athways. IL-33 is synthesized in response to cytokines such as IL-1β (but also TNFα) and plays an important role in T

2-associated immunology, by triggering an increase secretion of IL-5 and IL-13 from polarized T

2 cells. Overexpression of IL-33 in mice leads to severe pathological changes in mucosal organs.

General Information
In 2006, T. Waerner, et al. characterized ILEI as a key player in both epithelial to mesenchymal transition and late-stage carcinoma progression. EMT is a process that involves changes in gene expression, cytoskeleton organization, cell adhesion, and extracellular matrix composition. This process is important during tissue formation and organogenesis, but also during cancer progression, metastasis and chronic inflammation. ILEI is overexpressed and/or altered in intracellular localization in multiple human tumors, an event strongly correlated to invasion/EMT, metastasis formation, and survival in human colon and breast cancer.

General Information
The lymphocyte activation gene-3 (LAG-3, CD223), a member of the immunoglobulin superfamily (IgSF) related to CD4, binds to the major histocompatibility complex (MHC) class II molecules but with higher affinity than CD4 [1]. Several alternative mRNA splice-variants of human LAG-3 have been described, two of them encoding potential secreted forms: LAG-3V1 (i.e. the D1-D2 domains of the protein, 36 kDa) and LAG-3V3 (D1-D3, 52 kDa) [1,2]. The longer form was detected by ELISA in the serum of healthy individuals as well as of tuberculosis patients with a favorable outcome [3].
