Product Description
Inhibitor of intracellular protein transport and protein secretion, interfering with trafficking in the trans-Golgi network, leading to the accumulation of cycling proteins in ERGIC clusters. Blocks ADP-ribosylation factor (Arf) in an inactive GDP-bound conformation and thereby prevents binding of COPI coats to ERGIC and Golgi membranes. Upon brefeldin A treatment the Golgi rapidly tubulates and fuses with the ER by an energy-, temperature-, and microtubule-dependent process. In contrast, the drug has little effect on the ERGIC, which keeps its identity, although the ERGIC clusters become larger and more uniformly distributed in the cytoplasm of the cells. Inhibits intracellular collagen degradation. Also inhibits apical Na+ channels in epithelia. Induces apoptosis.

Product Description
The cytoskeleton-linking membrane protein of 62kDa (CLIMP-63) is a marker for the reticular subdomain of the rough endoplasmatic reticulum (ER) and is excluded from the outer nuclear membrane. Concentration of CLIMP-63 into patches may enhance microtubule binding on the cytosolic side and contribute to ER morphology by the formation of a protein scaffold in the lumen of the ER, linking the rough ER to microtubules.

Product Description
Giantin, the largest Golgi membrane protein, is C-terminally anchored to cis- and medial-Golgi membranes and forms a long ~250nm coiled-coil rod that directly interacts with microtubules in a cell-cycle regulated way. It is an excellent marker for mammalian golgi complex. Giantin plays also a role in docking COPI vesicles to Golgi membranes. It is speculated that after mitosis giantin aids in guiding vesiculated Golgi membranes to the microtubule organizing center to reform a new Golgi apparatus.

The transmembrane protein GPP130 is localized to the cis-Golgi, where it is targeted by virtue of its lumenal coiled-coil domain and mediates trafficking in the late endosome-bypass pathway. GPP130 continuously recycles between early Golgi and distal compartments.

General Information
The trans-Golgi network (TGN) is part of the secretory pathway of eukaryotic cells which is distinct from the Golgi stack.The TGN is important in the later stages of protein secretion where it seems to play a key role in the sorting and targeting of secreted proteins to the correct destination. Brefeldin A, Prod. No. ALX-350-019 causes the contents of the Golgi stack to be redistributed into the endoplasmatic reticulum while the TGN collapse upon the microtubule organizing center. This makes immunofluorescence detection of TGN38, following Brefeldin A treatment, a useful tool for the discrimination of the TGN from the Golgi stack.
