TRADD Protein Is an Essential Component of the RIG-like Helicase Antiviral Pathway: M.C. Michallet, et al.; Immunity 28, 651 (2008) Abstract
General Information
RIG-I (retinoic acid-inducible gene I; Ddx58) and MDA5 (melanoma differentiation-associated gene 5, also known as Ifih1 or Helicard) are proteins that sense viral replication intermediates, such as double-stranded RNA and triggers the host antiviral programs. These molecules signal the downstream activation of NF-κB and IFN regulatory factor (IRF) -3, which coordinately regulate the expression of type-I interferons.
Cardif (also called VISA/IPS-1/MAVS) is a new CARD (caspase activation and recruitment domain)-containing adaptor protein that interacts with the CARD domain of RIG-I and MDA5, leading to the activation of NF-κB and IRF3. Cardif is located to the mitochondrial outer membrane. Removal of the mitochondrial-targeting domain of cardif abolishes its ability to induce IFNs. Cardif is cleaved and inactivated by NS3-4A, a serine protease from hepatitis C virus known to block interferon-β production.

Characterization of three novel monoclonal antibodies against hepatitis C virus core protein: D. Moradpour, et al.; J. Med. Virol. 48, 234 (1996) Abstract
General Information
Hepatitis C virus (HCV) is the primary cause of non-A, non-B hepatitis and results in most HCV-infected people developing chronic infections, liver cirrhosis and hepatocellular carcinoma. The 21kDa core protein of HCV is well conserved among different HCV genotypes and may suppress hepatitis B virus replication in a phosphorylation dependent manner.
BACKGROUND/TECHNICAL INFORMATION Immunocytochemical staining of HCV core protein in transfected chimp hepatocytes yields a staining pattern consistent with cytoplasmic and vesicular staining.

General Information
MDA5 is a highly conserved helicase whose ATPase activity can be stimulated by RNA and less potently by DNA. It contains two N-terminal CARD domains and a C-terminal helicase domain. The helicase domain, once freed from the CARD domains, then translocates from the cytoplasm to the nucleus where it acts on chromatin architecture. This could allow a more easy access of CAD, thereby accelerating the degradation of genomic DNA.

General Information
Pin1, a member of the parvulin family of peptidyl-prolyl-cis-trans-isomerases (PPIases), specifically isomerizes only the phosphorylated  Ser/Thr-Pro bonds in certain proteins like Bcl-2, Cdc25C, Cdc27, p53 and Tau. Binding of Pin1 to p53 stimulates the DNA binding activity and transactivation function of p53. Pin1 also regulates the expression of cyclin D1 and is strikingly overexpressed in a subset of human tumors like breast cancer and oral squamous cell carcinoma.
