General Information
BACKGROUND/TECHNICAL INFORMATION For in vitro stimulation we recommend the following procedure:
1. Bone marrow-derived mouse dendritic cells (BM mDC) are cultured 8x104/100μl in RPMI containing
10% FCS over night at 5% CO2 and 37°C.
2. The medium is removed and replaced by 100μl OPTI MEM (Invitrogen) and subsequently
the transfection mixture is added: 25μl OPTI MEM containing 0.025 to 0.25 Poly(dA:dT) + 25μl OPTI MEM
containing 0.5μl Lipofectamine 2000 (Invitrogen) or alternative transfection agents.
3. Over night stimulation at 5% CO2 and 37°C.
4. Measurement of IFN-α/β in culture supernatants.

General Information
RIG-I (retinoic acid-inducible gene I; Ddx58) and Mda5 (melanoma differentiation-associated gene 5, also known as Ifih1 or Helicard) are proteins that sense viral replication intermediates, such as double-stranded RNA and triggers the host antiviral programs. These molecules signal the downstream activation of NF-κB and IFN regulatory factor (IRF) -3, which coordinately regulate the expression of type-I interferons.
Cardif (also called VISA/IPS-1/MAVS) is a new CARD (caspase activation and recruitment domain)-containing adaptor protein that interacts with the CARD domain of RIG-I and Mda5, leading to the activation of NF-κB and IRF3. Cardif is located to the mitochondrial outer membrane. Removal of the mitochondrial-targeting domain of cardif abolishes its ability to induce IFNs. Cardif is cleaved and inactivated by NS3-4A, a serine protease from hepatitis C virus known to block interferon-β production.

General Information
RIG-I (retinoic acid-inducible gene I; Ddx58), a member of the DEAD-box RNA helicases, is activated by cytoplasmic dsRNA produced during the viral replication. The protein is characterized by a N-terminal region with two caspase recruitment domains (CARD) and a C-terminal region harboring potential ATP-dependent RNA helicase activity. RIG-1 recruits the CARD adaptor inducing IFN-β (Cardif) in a CARD-CARD-dependent manner resulting in NF-κB and IRF3 activation.
